Ion Torrent (314 Chip)

UAGC offers Next Generation Sequencing on the Ion Torrent Personal Genome Machine. This platform allows parallel sequencing of small genomes, targeted re-sequencing, whole genome and exome validation, as well as RNA sequencing.

Sample Input

A variety of sample types can be run on the Ion Torrent, including; Genomic DNA, PCR Product and RNA

Expected Results

Researchers can choose the level of coverage best suited to their project:

  • 314 Chip: 20Mb Expected Results, 200bp average length
  • 316 Chip: 200Mb Expected Results, 200bp average length

Accuracy: >99.99% consensus and >99.6% raw

Sample Preparation Details

DNA sample requirements

  • 2µg total DNA required
  • DNA sample should be suspended in ≤ 130µL LowTE
  • DNA should not be degraded
  • DNA should have an OD 260/280 ratio ≥ 1.8
  • DNA must be double stranded
  • DNA should not be the result of whole genome amplification (or other process that can compromise accurate representation)
  • DNA should contain no particulate matter

RNA sample requirements

  • RNA should not be degraded
  • RNA integrity number (RIN) greater than 7

Whole transcriptome

  • 125-625ng of poly(A) RNA or 250-625ng of rRNA-depleted total RNA
  • Suspended in 10uL Nuclease-free water
  • Absent of contaminating rRNA
Workflow

How to Get Started with your Sample Submission: 

  • Login to Sample Lifecycle Manager (SLM) and select 'Ion Torrent' as your workflow.
  • Upload your sample information. Select relevant processing options. 
  • Click submit. 
  • Record and attach the 4 letter code at the end of your SLM submission to your samples and send or deliver to UAGC. 
 
Quality Control of Samples

Each Ion Torrent sample will go through the following quality control checks before it is processed  

  • Pico/RiboGreen Assay to verify the quantity of starting material
  • Bioanalyzer DNA/RNAchip to check the material quality

Customers will be asked for more DNA/RNA if their sample fails either of the above check points.

Sequencing is avaiable using the 316 Chip as well.
